Synthesis of fluorescent C-nucleoside: 2-aminopyridine

Abstract:
RNA plays a key role in many biological processes due to its various functions and structures. The sensitivity of fluorescence spectroscopy has advantages to characterize RNA. Since canonical nucleosides do not fluoresce, the designing and synthesis of fluorescent-modified nucleosides are necessary for biochemical and biophysical studies. A fluorescent pyrimidine analog, 2-aminopyridine, was developed in this project using Hannesian's asymmetric synthetic scheme. The newly synthesized nucleoside has excitation and emission wavelengths at about 230 and 360 nm, respectively.
