Exploring the effective length for circular RNA to form a g-quadruplex structure

Abstract:
Circular RNAs (circRNAs) are a unique class of single-stranded RNAs distinguished by a covalently closed lack free 5' and 3' ends. Using bioinformatics analysis, the Balaratnam group discovered the presence of G-quadruplex (G4) forming sequences in circular RNAs. The closed structures in circRNAs impose significant topological and spatial constraints on their folding, dynamics, and ability to adopt secondary structure. We investigated the minimum length requirements for circRNAs to form G4 structures by using the human CircRNA_DDX54_0018 as a model system. Using circular dichroism (CD) spectroscopy, we identified a length-dependent inhibition of G-quadruplex (G4) structure formation in circular RNAs shorter than 100 nucleotides. Our findings therefore establish a minimal length requirement for circRNAs to adopt a G4 conformation.
