Proximity labeling to identify binding partners of meiotic mRNA interceptor protein in Schizosaccharomyces pombe

Abstract:
Meiotic mRNA interceptor 1 (Mmi1) is a protein that regulates the transition between mitosis and meiosis in the fission yeast Schizosaccharomyces pombe. Mmi1 suppresses meiotic progression by binding to meiotic RNAs with its C-terminal YTH domain, which recognizes specific RNA sequences called determinants of selective removal (DSRs). It then recruits the nuclear exosome to degrade these transcripts via a poly(A)-dependent pathway, thus preventing entry into meiotic prophase 1. Mmi1 has also been implicated in chromatin remodeling by associating with the histone methyltransferase Clr4 at meiotic gene islands, thereby promoting chromatin condensation. Since Mmi1 does not have any known intrinsic catalytic functions, it likely achieves these diverse functions through interaction partners that regulate its functions in vivo. Here, I present my findings in characterizing Mmi1’s interaction network via proximity labeling with TurboID.
